Trabzanoflast

Trabzanoflast is a brain-penetrant NLRP3 inhibitor being developed to resolve neuroinflammation underlying neurologic conditions.

Trabzanoflast is a small molecule drug candidate that has been discovered and engineered by Rezera to achieve a new benchmark for brain penetration.

Trabzanoflast (NT-0150) is Rezera’s proprietary next-generation, brain-penetrant NLRP3 inhibitor. Its chemical structure was specifically selected for potent and selective NLRP3 inhibition and optimized to achieve high brain penetration. In preclinical studies, trabzanoflast has demonstrated robust efficacy, reducing central and peripheral inflammatory markers across multiple animal models.

Trabzanoflast has completed a Phase 1 clinical trial to assess the safety and tolerability of trabzanoflast, as well as its pharmacokinetics and biodistribution in healthy volunteers. The topline results are expected in Q4 of 2026.

Trabzanoflast has been designed to achieve robust brain exposure, with the goal of enabling NLRP3 inhibition in the CNS and addressing neuroinflammatory disorders.

The role of chronic inflammation in the central nervous system, including the brain, eye, ear, and spinal cord, is central to disease initiation and progression. The challenges to effectively address chronic inflammation in these compartments have been two-fold. First, these compartments have historically been difficult to reach at high levels, even for small molecules.

The second challenge is delivering a chronic therapy while preserving other immune pathways involved in natural responses to infection. We have designed trabzanoflast to selectively inhibit NLRP3, with the goal of preserving these other immune pathways and allowing patients to respond naturally to infection.
