Ruvonoflast
Ruvonoflast is a potent oral NLRP3 inhibitor that has been studied in over 400 patients.
Ruvonoflast has been engineered to optimize for both tissue and brain penetration.
Ruvonoflast inhibits NLRP3. Across several clinical trials, we have observed reductions in downstream pro-inflammatory cytokines, including IL-1β IL-18, and IL-6, and in biomarkers including fibrinogen, hsCRP, and Lp(a). Across our four completed, placebo-controlled clinical trials to date, we have observed no increased rate of infections among participants treated with ruvonoflast compared with a placebo.
Ruvonoflast is an investigational oral NLRP3 inhibitor advancing to Phase 3 development in peripheral arterial disease (PAD). The program is designed to address residual inflammatory risk by targeting a powerful, upstream driver of chronic inflammation.
As ruvonoflast is designed to achieve broad tissue distribution, including penetration into the central nervous system, we believe that this makes it well suited to target chronic inflammation in the compartments where inflammation-driven damage occurs. In cardiovascular diseases such as peripheral artery disease, this means the potential for activity in the arterial wall and plaque, as well as in small blood vessels and skeletal muscles. Since most cardiovascular diseases also have a component of cerebrovascular disease, we believe that the high brain penetration of ruvonoflast is an additional positive.
We believe these attributes also make ruvonoflast well suited for development in a variety of additional indications within and beyond cardiovascular, where high tissue penetration is key to reducing the damage caused by chronic inflammation. We expect to announce additional indications for ruvonoflast development in the future.
